Program Overview

This study is testing a combination of two treatments, SX-682 and atezolizumab, for non-small cell lung cancer (NSCLC) that has spread to other parts of the body or has come back after treatment. SX-682 works by blocking proteins that might help the immune system kill cancer cells. Atezolizumab is an immunotherapy that helps the immune system attack cancer and stop it from growing. The goal is to see if this combination is effective for treating advanced or returning NSCLC.

Description

In this study, patients will take SX-682 by mouth twice a day and receive atezolizumab as an injection once every 21 days. This treatment cycle can continue for up to 5 years unless the cancer progresses or side effects become too severe. Patients will have regular scans and tests, including MRI and CT scans, blood tests, and possibly tumor biopsies, to monitor their response to the treatment.

After finishing the study treatment, patients will have follow-up visits within 30 days if they stop treatment due to safety concerns, cancer progression, or early withdrawal. They will then have check-ups every 12 weeks for up to 7 years to monitor their health.

Eligibility Criteria

Inclusion Criteria

  • Must be 18 years or older.
  • Must understand and agree to sign a consent form.
  • Must have non-small cell lung cancer without certain genetic mutations.
  • Cancer must have spread or returned after previous treatments.
  • Must have tried and not responded to previous immunotherapy and chemotherapy.
  • Must have at least one measurable tumor.
  • Must be in good overall health with a performance status of 0 or 1.
  • Must provide a tumor sample or undergo a biopsy if needed.
  • Must have adequate blood counts and organ function.
  • Must agree to use effective birth control during and after the study.
  • Women must have a negative pregnancy test before starting treatment.

Exclusion Criteria

  • Have another active cancer within the last 2 years.
  • Have symptomatic brain metastases or require steroids for brain metastases.
  • Have untreated brain metastases unless they meet specific criteria.
  • Have active autoimmune diseases requiring treatment in the past year.
  • Cannot stop steroid treatment before starting the study.
  • Have a history of organ transplant or stem cell transplant requiring immunosuppressives.
  • Have current or past severe lung inflammation.
  • Have had severe side effects from previous immunotherapy.
  • Have had recent radiation or major surgery.
  • Have heart or lung conditions that are not well-controlled.
  • Have active infections like hepatitis or tuberculosis.
  • Have uncontrolled HIV infection.
  • Have any condition that would prevent completing the study.
  • Have received a live vaccine recently or plan to during the study.
  • Have a history of severe allergic reactions to similar treatments.
