Study on Crizotinib for Treating Cancers with MET Exon 14 Deletion Genetic Changes

Program Overview
The purpose of this study is to see if crizotinib can effectively treat cancers with a genetic change known as MET exon 14 deletion. Crizotinib belongs to a group of drugs called tyrosine kinase inhibitors, which work by blocking enzymes that help cancer cells grow and spread. It may also stop the growth of new blood vessels that tumors need to grow.
Description
The main goal of this study is to find out how many patients experience a reduction in their cancer with crizotinib treatment. Researchers will also look at how many patients are alive and have no disease progression after six months of treatment. They will study the time until the cancer progresses or the patient passes away. Additionally, the study will explore other potential markers that might predict how well the treatment works or why it might not work, using various tests on tumor samples and imaging scans.
Patients in the study will take crizotinib by mouth twice a day in 28-day cycles. These cycles will continue as long as the cancer does not progress and the patient can tolerate the treatment. During the study, patients will have tumor biopsies, imaging scans, and blood tests to monitor their response to the treatment.
After finishing the study treatment, patients will have follow-up visits every three months for two years, and then every six months in the third year.
Eligibility Criteria
Inclusion Criteria
- Must meet the eligibility criteria of the main MATCH study before joining this sub-study.
- Must have a specific genetic change called MET exon 14 skipping or other approved mutations.
- Must have a recent ECG test showing no major heart rhythm problems.
- Must not be allergic to crizotinib or similar drugs.
- Must not have taken certain other cancer drugs before.
- Must not have severe lung conditions or a history of certain lung diseases.
- Must not have had recent major heart problems or certain gastrointestinal issues.
- Must not be taking certain medications that affect how crizotinib works.
- Must not have had major surgery recently.
