This study is exploring a new treatment for advanced solid cancers using a drug called ETX-19477. The drug is designed to target a specific weakness in cancer cells, which can help stop their growth. The study will be conducted in two parts to determine the best dose and to evaluate the safety and effectiveness of the treatment.
Description
Cancer cells often have a problem called replication stress, where their DNA gets damaged during the process of copying itself. This can lead to the cells stopping their growth or dying. The drug ETX-19477 works by blocking a protein called PARG, which plays a role in repairing this DNA damage. By blocking PARG, the drug aims to make it harder for cancer cells to fix themselves, potentially stopping their growth or causing them to die. This study will test if ETX-19477 can effectively target this weakness in cancer cells and help treat advanced solid cancers.
Eligibility Criteria
Inclusion Criteria
- Adults aged 18 years or older.
- Have a confirmed diagnosis of advanced solid cancer that cannot be cured or removed by surgery, excluding brain tumors.
- Cancer must have worsened after previous treatments or the patient cannot tolerate standard treatments.
- Must not have taken any experimental drugs within a few weeks before starting the study.
- Expected to live at least 3 more months.
Exclusion Criteria
- Taking more than 10 mg of prednisone or similar steroids daily.
- Recent radiation therapy within a few weeks before starting the study.
- Untreated or worsening brain cancer.
- Digestive issues that could affect the absorption of the study drug.
- Heart problems or a family history of heart issues.
- Active infections like hepatitis or HIV, unless well-controlled.
- Serious kidney, pancreas, or liver problems.
- Other cancers needing treatment within the last 2 years, except for certain skin or bladder cancers.
- Taking certain medications that could interfere with the study drug.
