Program Overview

This study is investigating a new method to choose the best treatment for patients with metastatic breast cancer that is positive for estrogen receptors and has not responded to aromatase inhibitors and CDK4/6 inhibitors. Researchers are using the biology of the tumor to predict which treatment might work best. The goal is to see if these predictions are accurate and to gather more information about the tumors to help design better treatments in the future.

Eligibility Criteria

Inclusion Criteria

  • Diagnosed with breast cancer.
  • Cancer has spread or cannot be cured.
  • Previously treated with anastrozole or letrozole and a CDK4/6 inhibitor.
  • Cancer has progressed while on or within 6 months of stopping the CDK4/6 inhibitor.
  • Have at least one tumor that can be biopsied, which is not just a hard bone lesion.
  • Have a performance status score of 0 to 2, indicating full activity or some restriction in strenuous activities.
  • Aged 18 or older.
  • Able to understand and sign a consent form agreeing to participate in the study.

Exclusion Criteria

  • Have received treatment with fulvestrant, alpelisib, everolimus, or capecitabine in the past 2 years.
  • Have another serious illness besides breast cancer with a life expectancy of less than 2 years.
  • Have another type of cancer that will need treatment within 2 years.
  • Have a low platelet count (less than 100,000/microliter).
  • Have a blood clotting time (INR) greater than 1.5.
